LABORATORY PROCEDURES FOR PLASMIDS REFERENCE NO: PP/2003/03/02 TITLE: QUALITY CONTROL CHECK OF PLASMID DNA INTRODUCTION This document describes the basic procedure to be followed when checking the quality of the plasmid DNA upon arrival of a new plasmid for the public collection. PROCEDURE
b) Analysis of the restriction
site pattern by agarose gel electrophoresis after single and (when possible)
double restriction enzyme digestion.
The plasmid size estimated from the molecular weight of the ccc DNA
must match the depositor's data. - The plasmid size estimated from linearized DNA must match the
depositor's data. - Where two or more restriction sites are known, the presence and the
